چکیده

The superoxide dismutase (dismutase) which has already been found to be useful in detecting the role of O2 ain certain enzymic reactions has here been shown to be applicable to the study of nonenzymic reactions, as well. The aerobic oxidation of sulfite, which proceeds by a free radical chain mechanism, has been investigated with the aid of superoxide dismutase. The “spontaneous” oxidation of sulfite, which is catalyzed by traces of heavy metals, was unaffected by superoxide dismutase in the absence of ethylenediaminetetraacetate yet was inhibited by 1 ng per ml levels of the dismutase in the presence of EDTA. Dimethyl sulfoxide (DMSO) has been found to be a powerful catalyst of the aerobic oxidation of sulfite. This is a complex reaction with a mechanism which appears to change with changes in the relative concentrations of sulfite and of DMSO. When the ratio of [sulfite] to [DMSO] was high, sulfite oxidation was inhibited by dismutase. In contrast, when the ratio of [sulfite] to [DMSO] was low, dismutase stimulated the oxidation of sulfite. The effects of variation of the concentrationn of sulfite, DMSO, and oxygen on the DMSO-induced sulfite oxidation have been explored. DMSO was found to contain approximately 0.2% of an impurity which was a very effective radical scavenger. This impurity could not be eliminated by treatment with charcoal and vacuum distillation, but it could be destroyed by treatment with Hz02. The reduction of cytochrome c by sulfite and the effects of EDTA, DMSO, oxygen, and dismutase on this reduction have also been studied. The results, which have permitted clarifying deductions concerning these very complex reactions, could not have been achieved, were the superoxide dismutase not available.

Oxidation of nicotinamide coenzyme dimers by one-electron-accepting proteins.

The nicotinamide nucleotide dimers (NAD)2 and (NADP)2, obtained by electrochemical reduction of NAD+ and NADP+, are able to reduce such single-electron acceptors as the proteins cytochrome c, azurin and methaemoglobin, though at different rates. Under the same conditions the reduced nicotinamide coenzymes NADH and NADPH are not able to reduce these proteins at measurable rates unless a catalyst...

Comparative modelling of 3D-structure of Geobacter sp. M21 (a metal reducing bacteria) Mn-Fe superoxide dismutase and its binding properties with bisphenol-A, aminotriazole and ethylene-diurea

Superoxide dismutase play important roles in iron-respiratory bacteria such as Geobacteraceae as an antioxidant defense, and probably an effective enzyme of electron transfer network. Regarding the application of iron-respiratory bacteria in environmental biotechnology particularly biodegradation and bioremediation, understanding the mechanism of inhibition/induction of superoxide dismutase by ...

Antioxidant and Free Radical Scavenging Potential of Yakuchinone B Derivatives in Reduction of Lipofuscin Formation Using H2O2-Treated Neuroblastoma Cells

Background: The progressive accumulation of misfolded and aggregated proteins in neurons is an accepted mechanism in aging. Overproduction of reactive oxygen species (ROS), referred to as oxidative stress, is currently believed to play a pivotal role in this process. Lipofuscin as a histological index of aging results from cross-links between oxidized proteins and lipids. Therefore, to attenuat...

Redox properties of the lipocalin alpha1-microglobulin: reduction of cytochrome c, hemoglobin, and free iron.

alpha1-Microglobulin (alpha1m) is a 26-kDa plasma and tissue glycoprotein. The protein has a heterogeneous yellow-brown chromophore consisting of small unidentified prosthetic groups localized to a free thiol group (C34) and three lysyl residues (K92, K118, and K130) around the entrance to a hydrophobic pocket. It was recently reported that alpha1m can bind heme and that a C-terminally processe...

Superoxide dismutase-inhibitible reduction of cytochrome c by the alloxan radical. Implications for alloxan cytotoxicity.

Cytochrome c was reduced when superoxide was generated from xanthine oxidase in the presence of alloxan, and by the reaction of alloxan and with reduced glutathione. In each case, most of the reduction was inhibited by superoxide dismutase, but considerably more enzyme was required than with superoxide alone. This indicates that the superoxide dismutase-inhibitible cytochrome c reduction was ma...
